تاثیر داروهای ضدقارچ آزول بر تولید سایتوکینهای التهابی در کراتینوسیتهای انسانی

چکیده
مقدمه: اگر چه داروهای ضد قارچ به صورت موفقیتآمیزی در درمان عفونتهای قارچی به کار میروند، لیکن اخیراً گزارشهایی مبنی برنقش برخی از آنها در القا و یا سرکوب پاسخ های سیستم ایمنی ارایه شده است. با توجه به نقش سیستم ایمنی در پاسخ به عوامل عفونی هرگونه تغییر در طرح واره سایتوکینی وضعیت پاسخگویی فرد را تحت تاثیر قرار خواهد داد.
هدف: بررسی اثر داروهای ضد قارچ آزول بر تولید سایتوکینهای التهابی از کراتینوسیتهای انسانی.
مواد و روشها: در یک مطالعه آزمایشگاهی (In vitro) ، سلولهای اولیه کراتینوسیت کشت داده شده و با غلظتهای مختلف کتوکونازول، فلوکونازول و گریزئوفولوین تیمار شدند. سپس میزان IL-1 وTNF-α ترشح شده توسط سلولهای کراتینوسیت در محیط کشت توسط روش Quantitative Enzyme Immunoassay و میزان بیان ژنهای کدکننده سایتوکینهای التهابی با روش Real-Time PCR سنجیده شد. دادها با استفاده از آنالیز واریانس یک طرفه One Way ANOVA تجزیه و تحلیل شد.
نتایج: تیمار سلولها با غلظتهای مختلف فلوکونازول( 3/3 ، 10 و 30 میکروگرم بر میلیلیتر) و غلظت پایین کتوکونازول باعث کاهش معنی دار ترشح IL-1 شد (P<0.001 )، اما گریزئوفولوین در غلظتهای مشابه چنین تاثیری را از خود نشان نداد. همچنین غلظتهای مختلف ترکیبات مذکور در ترشح TNF-α تاثیری نداشت. بررسی کمی بیان ژن کدکننده IL-1 نشان داد که نسخهبرداری از ژنهای مذکور به دنبال تیمار سلولها با فلوکونازول و کتوکونازول کاهش مییابد.
نتیجهگیری: ترکیبات ضد قارچ آزول ممکن است با سازوکارهای مختلف موجب تعدیل بیان و ترشح سایتوکینها شده و بدین ترتیب آثار خود را در جهتدهی پاسخهای ایمنی توسط سلولهای کراتینوسیت اعمال نمایند.
Abstract
Introduction: Antifungal drugs have been successfully used in treatment of fungal infections. Recently, immunomodulatory effects of some of these agents have been reported. Base on immune system role in the treatment of various infection, alteration in cytokine pattern would be influenced the immune responsiveness.
Objective: Study the expression and secretion of the inflammatory cytokines in human keratinocyte.
Materials and Methods: In this invitro study,cultured keratinocytes were treated with different concentrations of Fluconazole, Ketoconazole and Griseofulvin. The level of IL-1 and TNF-α by keratinocytes in cultured supernatant were measured by Quantitative Enzyme Immunoassay technique and their expression were evaluated by using real time PCR. Data was analysed with one way ANOVA test(Varyance).
Results: Treatment of keratinocytes with different concentrations of Fluconazole (3.3,10,30 µ/ml) and low concentration of ketoconazole caused to decrease of IL-1 secretion (P<0.001), but Griseofulvin did not show this effect at the same concentrations. In addition, the examined drugs had no effect on TNF-α secretion. Quantitative analysis of IL-1 encoding genes revealed that transcription on these genes might be suppressed following treatment with Fluconazole or ketoconazole.
Conclusion: Antifungal azoles might be modulated cytokines expression and secretion as well as affect the direction of immune response induced by keratinocytes.
