اثر مصرف خوراکی میوه گیاه خارخاسک بر یادگیری و حافظه موش صحرایی دیابتی: تعیین نقش پراکسیداسیون لیپیدی

مقدمه: دیابت در دراز مدت با اختلال یادگیری و حافظه همراه میشود. با توجه به اثر ضد دیابتی خارخاسک و اثر آنتیاکسیدانی آن در این پژوهش تأثیر این گیاه بر یادگیری و حافظه موشهای صحرایی دیابتی بررسی شد. هدف: بررسی تأثیرگیاه خارخاسک بر یادگیری و حافظه موشهای صحرایی دیابتی مواد و روشها: 32 موش صحرایی به چهار گروه کنترل، کنترل تحت تیمار با خارخاسک، دیابتی، و دیابتی تحت درمان با خارخاسک تقسیم شدند. برای بررسی حافظه و یادگیری حیوانات، عملکرد در آزمون اجتنابی غیر فعال و درصد رفتار تناوب با استفاده از ماز Y تعیین شد. در هموژنه بافت هیپوکامپ مغز حیوانات، میزان مالون دیآلدئید تعیین شد. نتایج: در موشهای گروه دیابتی (01/0>p) در پایان کار در حین عبور کاهش تأخیر بروز کرد و گروه دیابتی تحت تیمار با گیاه، افزایش معنیدار این پارامتر را در مقایسه با گروه دیابتی نشان داد(01/0>p). درصد تناوب نیز در جانداران دیابتی بطور معنیدار کمتر از گروه کنترل بود(05/0>p) و درصد تناوب در گروه دیابتی تحت تیمار افزایش معنیدار در مقایسه با گروه دیابتی نشان داد(05/0p<). بهعلاوه، موشهای دیابتی افزایش معنیدار در سطح بافتی مالون دیآلدئید(01/0>p) نشان دادند یعنی درمان با خارخاسک میزان مالون دیآلدئید(01/0>p) را به صورت معنیدار کاهش داد. نتیجهگیری: تجویز خوراکی خارخاسک بر توانایی نگهداری اطلاعات در انبار حافظه و یادآوری آنها در حیوانات دیابتی تأثیر دارد و موجب بهبود حافظه فضائی حیوانات دیابتی میشود. بخشی از آثار سودمند گیاه با کاهش پراکسیداسیون لیپیدی در بافت هیپوکامپ اعمال میشود.
Introduction: Diabetes in long term is associated with learning and memory deficits. Given the Anti-diabetic and antioxidant activities of Tribulus terrestris (TT). Objective: to evaluate the effect of oral administration of Tribulus terrestris on learning and memory in diabetic rats. Materials and Methods: Thirty two Wistar rats were divided into control, TT-treated control, diabetic, and TT-treated diabetic groups. To evaluate their learning and memory, initial (IL) and step-through latencies (STL) were determined at the end of study using passive avoidance test and alternation behavior percentage was obtained using Y maze. Level of malondialdehyde in hippocampal homogenate was also measured at the end of the study. Results: STL significantly decreased in diabetic group (p<0.01) and TT-treated diabetic group showed a significantly higher STL (p<0.01), compared to the diabetic counterparts. Alternation percentage was significantly lower in the diabetic group relative to the controls (p<0.01) and treated diabetic group did show a significant difference in comparison with the diabetic group (p<0.05). Diabetic rats also exhibited a significant increase in tissue level of malondialdehyde (p<0.01) and TT treatment significantly reduced MDA level (p<0.01) in the diabetic group. Conclusion: TT treatment could enhance the capability of consolidation and recall in diabetic animals and could also improve spatial memory in them, partly via attenuation of lipid peroxidation in the hippocampus.
