چکیده
مقدمه: دیابت تیپI یکی از بیماریهای خودایمن است که با تخریب سلولهای بتای جزایر لانگرهانس همراه است و بیشتر افراد را، در نوجوانی و جوانی مبتلا میکند. زمینههای ژنتیکی، محیطی و نیز ایمونولوژی در ایجاد و پیدایش عوارض مختلف این بیماری نقش دارند. TGFβ یکی از سیتوکینهایی است که در تنظیم و مهار پاسخهای ایمنی نقش دارد و ترشح غیرطبیعی آن در بیماریهای خودایمنی دیده میشود. پلی مرفیسم ناحیه 915+ ژن TGFβ، تولید و ترشح این سیتوکین را تحت تاثیر قرار میدهد و بهعنوان عامل خطر در بیماریهای خودایمن قلمداد می شود.
هدف: مقایسه پلی مورفیسم کدون 25 ژن TGFβ در بیماران با گروه شاهد.
مواد و روشها: این مطالعه از نوع مورد – شاهدی بود و 75 بیمار دچار دیابت تیپ I که تشخیص آنها حداقل 2 سال پیش از نمونهگیری توسط پزشک متخصص قطعی شده و با انسولین درمان میشدند و گروه کنترل از داوطلبان اهدای خون که به سازمان انتقال خون ایران مراجعه کرده بودند پس از اخذ رضایت انتخاب شدند.
از نرم افزار SPSS برای تجزیه و تحلیل دادهها و از آزمون آماری مربع کای با حدود اطمینان95% برای مقایسه نتایج استفاده شد .
نتایج: تفاوت آماری معنیداری در پلیمرفیسمC/G در ناحیه 915+ بین بیماران و گروه شاهد وجود نداشت.
بحث و نتیجهگیری: اگر چه پلیمرفیسم ناحیه 915+ ژن TGF-β تولید این سیتوکین را تحت تاثیر قرار میدهد و این سیتوکین در تنظیم پاسخهای ایمنی نقشاساسی دارد، اما وجود نوکلیوتیدهای C یا G در جایگاه 915+ کدون 25 نمیتواند بهعنوان فاکتور خطر یا مستعدکننده در بروز بیماری دیابت درنظرگرفتهشود.
Abstract
Introduction: Type 1 diabetes is an autoimmune disease which is characterized by T-cell mediated destruction of pancreatic β-cells. A variety of environmental and genetic factors are involved in the development of the disease.
Transforming Growth Factor Beta-1 (TGFβ) is a multifunctional cytokine that affected regulation and difference of immune responses and there is abnormal secretion of this auto immune disease.
Genetic polymorphisms in the TGFβ gene influenced production and secretion of cytokine and figured as a risk factor in auto immune disease.
Objective: In this study, single nucleotide polymorphism of TGFβ at codon 25 is investigated in type 1 diabetic patients and compared with healthy controls.
Materials and Methods: This was a case control study which 75 type 1 diabetic patients who had definitely diagnosed at least 2 years before sampling and were under Insulin therapy were selected. Control group were selected from voluntarily blood donors who had referred to IRAN Blood Transfusion Organization.
Data were analyzed by SPSS and using chi- square test with 95% confidence interval.
Results: The findings showed that there were not statistically significant differences in G C polymorphism at +915 between cases and control groups (p value 0.05).
Conclusion Although TGFβ polymorphism at +915 regions affects the production of cytokine regulates immune responses this study showed that single nucleotide polymorphism in this region can not be involved in onset of type 1diabetes and can not be considered as risk factor.
